چکیده

Liver cancer is one of the most common digestive malignancies. Conventional therapies such as surgical resection and chemotherapy are ineffective, and it is rare to achieve complete remission or cure. Application of RNA interference (RNAi) to reduce oncogene expression and thereby change the biology of tumor cells is one of the most important methods in tumor therapy (Sibley et al., 2010). Livin and Survivin are members of the Inhibitor of Apoptosis (IAP) family of proteins, and are highly expressed in hepatocellular carcinoma cells. These two genes exert their anti-apoptotic effects through the IAP repeat domains (baculovirus-IAP repeat, BIR). Knockdown of either Livin (Liu et al., 2010) or survin (Hung et al., 2012) can manifest therapeutic effects against liver cancer. It has also been shown that these two genes, together with XIAP, exert a synergistic effect that serves to inhibit cellular proliferation and to promote cell apoptosis in human bladder cancer cells (Yang et al., 2010) and human colon cancer cells (Oh, 2011; Carrasco et al., 2011). However, it remains unknown as to whether

Effects of shRNA-silenced livin and survivin on lung cancer cell proliferation and apoptosis.

PURPOSE To evaluate the effects of short hairpin RNA (shRNA)-mediated silencing of livin and survivin on the proliferation and apoptosis of A549 lung cancer cells. METHODS We designed and constructed the eukaryotic expression vectors pSilencer-livin and pSilencer-survivin which contain the Livin and Survivin genes, respectively, and transfected them into liposome-combined lung cancer A549 cel...

Effects of RNA interference-mediated knockdown of livin and survivin using monomethoxypolyethylene glycol-chitosan nanoparticles in MG-63 osteosarcoma cells.

MG-63 human osteosarcoma cells were transfected with short hairpin RNA (shRNA) against livin and survivin using monomethoxypolyethylene glycol‑chitosan (mPEG‑CS) nanoparticles (NPs) as carriers, with the aim of evaluating the effect on cell proliferation and apoptosis. mPEG‑CS NPs sized ~100 nm were prepared by ionic crosslinking. mPEG‑CS‑livin shRNA, mPEG‑CS‑survivin shRNA and mPEG‑CS‑(livin s...

Natural Killer Cell Cytotoxicity Against SKOV3 after HLA-G Downregulation by shRNA

Background: HLA-G is a nonclassical HLA class I molecule which, when elevated in tumor cells, is one of the main factors involved in tumor evasion of immune responses including NK and T cells. Objective: To evaluate the effect of HLA-G downregulation on NK cell cytotoxicity in tumor cell lines. Methods: The expression level of HLA-G was measured by real-time PCR and flowcytometry after transfec...

Suicide gene therapy for hepatocellular carcinoma cells by survivin promoter-driven expression of the herpes simplex virus thymidine kinase gene

The aim of this study was to investigate the selective killing effect of the herpes simplex virus-thymidine kinase/ganciclovir (TK/GCV) suicide gene system controlled by the survivin promoter on hepatocellular carcinoma (HCC) cells in vitro. Recombinant plasmid vectors driven by the survivin promoter were constructed. HepG2 HCC and LO2 normal huma...

STUDY OF HMGA2 GENE INHIBITION WITH SPECIFIC SHRNA AND SIRNA AND INVESTIGATION OF CORRESPONDING EFFECTS ON DOWNSTREAM GENE EXPRESSION IN MDA-MB-231 CANCER CELLS: A BIOINFORMATIC AND EXPERIMENTAL STUDY

Background & Aims: The use of siRNA to silence gene expression is increasingly expanding today. The aim of this study is to bioinformatically and experimentally investigate the inhibition of the HMGA2 gene and its corresponding effects on downstream genes expression rate in MDA-MB-231 cancer cell treated by shRNA and siRNA specific to HMGA2. Materials & Methods: To perform this bioinformatic a...
